基于PI3K/Akt信号通路探讨紫正地黄汤对人呼吸道合胞病毒感染小鼠肺损伤的作用机制*

摘要:目的:通过磷酯酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)信号通路,研究紫正地黄汤对呼吸道合胞病毒(RSV)感染小鼠肺组织炎症水平及病毒表达的影响,探讨紫正地黄汤对RSV感染小鼠肺组织可能发挥的抗炎抗病毒作用。方法:将小鼠随机分为空白组、模型组、对照组(利巴韦林组)及紫正地黄汤低剂量组、中剂量组、高剂量组。给药后对小鼠肺组织进行取材,计算小鼠肺指数;苏木精-伊红染色法(HE)染色后观察小鼠肺组织病理形态;酶联免疫吸附试验(ELISA)检测肺组织白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平;蛋白质免疫印迹(WB)检测PI3K、Akt及p-PI3K及p-Akt蛋白表达水平;定量聚合酶链式反应(q-PCR)检测RSV相对表达量;同时免疫荧光观察小鼠肺组织中RSV蛋白荧光强度。结果:较模型组,给药组小鼠肺组织炎性浸润得到有效改善,IL-6、TNF-α、IL-1β表达显著下调(P<0.05),p-PI3K及p-Akt蛋白表达明显提高(P<0.05),病毒含量减少(P<0.05)。结论:紫正地黄汤能有效降低RSV感染小鼠肺组织中病毒复制水平,且可上调p-PI3K及p-Akt蛋白的表达,并调控TNF-ɑ、IL-6、IL-1β等炎症因子的表达,减轻小鼠肺组织炎症程度,从而发挥抗炎抗病毒作用,减轻小鼠肺损伤。

Abstract:
Objective: To study the effects of Zizheng Dihuang decoction (ZZDHD) on on the level of lung tissue inflammation and viral expression in mice infected with Respiratory Syncytial Virus (RSV) through the phosphoinositide 3-kinase(PI3K)/protein kinase B (Akt), and to explore its potential anti-inflammatory and antiviral effects against RSV-induced lung injury in mice. Methods: Mice were randomly divided into blank group, model group, control group (ribavirin group), ZZDHD low concentration group, ZZDHD medium concentration group and ZZDHD high concentration group. The lung tissue of mice was sampled after administration and the lung index was calculated. The pathological morphology of lung tissue was observed after hematoxylin-eosin staining (HE) staining. Enzyme linked immunosorbent assay (ELISA) was used to detect the levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β). Protein expression levels of PI3K, Akt, phosphorylated PI3K (p-PI3K) and phosphorylated Akt (p-Akt) were detected by Western blotting (WB). Quantitative polymerase chain reaction (q-PCR) was used to detect the relative expression of RSV. Meanwhile, the RSV protein fluorescence intensity in mouse lung tissue was observed by immunofluorescence. Results: Compared with the model group, the administration groups showed significant improvement in inflammatory infiltration in lung tissue, significant downregulation of IL-6, TNF-α and IL-1β expression (P<0.05), significantly increased protein expression of p-PI3K and p-Akt (P<0.05), and reduced virus content (P<0.05). Conclusion: Zizhong Dihuang decoction can effectively reduce the virus replication level in the lung tissue of mice infected with RSV, up-regulate the expression of p-PI3k and p-Akt protein, regulate the expression of TNF-ɑ, IL-6 and IL-1β, and reduce the degree of inflammation in the lung tissue of mice, thus playing an anti-inflammatory and antiviral role and reducing the degree of lung injury in mice.
