甘草地上部分不同提取纯化部位HPLC指纹图谱及抗慢性非细菌性前列腺炎谱效关系研究*

摘要:

目的:建立甘草地上部分不同提取纯化部位的高效液相色谱(HPLC)指纹图谱,并探讨甘草地上部分化学成分与治疗慢性非细菌性前列腺炎功效的谱效关系。方法:以甘草地上部分不同提取纯化部位建立指纹图谱;以大鼠的生化指标及前列腺指数作为药效指标,考察甘草地上部分不同提取纯化部位治疗慢性非细菌性前列腺炎的功效。结果:确定了27个共有峰,指认了5个共有峰,分别为夏佛塔苷(17号峰)、异槲皮苷(22号峰)、紫云英苷(24号峰)、异甘草苷(25号峰)、槲皮素(26号峰);灰色关联度分析发现,所有共有峰与治疗慢性非细菌性前列腺炎作用的关联度均大于0.6,呈相关性;偏最小二乘分析发现,1号峰、2号峰、16号峰、19号峰、24号峰、25号峰对治疗慢性非细菌性前列腺炎作用的影响较大。结论:建立了甘草地上部分不同提取纯化部位的HPLC纹图谱,并指认了5个共有峰成分;16号峰、19号峰、24号峰(紫云英苷)、25号峰(异甘草苷)所代表的化学成分可能是甘草地上部分治疗慢性非细菌性前列腺炎的药效物质。

Abstract:

Objective: To establish high-performance liquid chromatography (HPLC) fingerprint profiles of different extracted and purified fractions from the aerial parts of Glycyrrhiza and explore the spectrum-effect relationship between their chemical components and the efficacy against chronic non-bacterial prostatitis (CNP). Methods: HPLC fingerprint profiles of different extracted and purified fractions from the aerial parts of Glycyrrhiza were established. Biochemical indicators and prostate index in rats were used as pharmacological indices to evaluate the anti-CNP efficacy of these fractions. Results: The established fingerprint profiles identified 27 common peaks, with five peaks specifically assigned as schaftoside (peak 17), isoquercitrin (peak 22), astragalin (peak 24), isoliquiritin (peak 25), and quercetin (peak 26). Grey correlation analysis revealed that all common peaks exhibited correlations (association degree>0.6) with the therapeutic effects on CNP. Partial least squares analysis indicated that peaks 1, 2, 16, 19, 24, and 25 had significant impacts on CNP treatment. Conclusion: HPLC fingerprint profiles of different extracted and purified fractions from the aerial parts of licorice were successfully established, with five characteristic components identified, and the components represented by peak 16, peak 19, peak 24 (astragalin ), and peak 25 (isoliquiritin) are likely the pharmacodynamic substances of the aerial parts of Glycyrrhiza in the treatment of chronic non-bacterial prostatitis.
