中药单体调控NF-κB通路干预胰腺癌研究进展*

摘要:综述核因子κB(NF-κB)信号通路在胰腺癌发生发展中的作用,并总结中药单体通过调控该通路干预胰腺癌的机制。NF-κB通路在胰腺癌的炎症微环境、细胞增殖、细胞凋亡、细胞侵袭、转移及化疗耐药中持续激活,是其关键致病机制之一。NF-κB通路是中药单体治疗胰腺癌的重要靶点,其多成分、多通路协同作用机制为胰腺癌的治疗策略优化提供了理论依据。中药单体可通过抑制NF-κB通路的活化、核转位及下游靶基因表达,诱导肿瘤细胞凋亡与周期阻滞,抑制肿瘤细胞增殖与侵袭,逆转上皮-间充质转化(EMT)及化疗耐药,调节肿瘤免疫微环境。

Abstract:This article reviews the role of the nuclear factor-kappa B (NF-κB) signaling pathway in the occurrence and development of pancreatic cancer, and summarizes the mechanisms by which traditional Chinese medicine (TCM) monomers intervene in pancreatic cancer by regulating this pathway. Persistent activation of the NF-κB pathway in the inflammatory microenvironment, cell proliferation, apoptosis, invasion, metastasis, and chemoresistance of pancreatic cancer is one of its key pathogenic mechanisms. The NF-κB pathway serves as an important target for TCM monomers in treating pancreatic cancer. Their multi-component, multi-pathway synergistic action mechanism provides a theoretical basis for optimizing treatment strategies for pancreatic cancer. TCM monomers can induce tumor cell apoptosis and cell cycle arrest, inhibit tumor cell proliferation and invasion, reverse epithelial-mesenchymal transition (EMT) and chemoresistance, and regulate the tumor immune microenvironment by inhibiting the activation, nuclear translocation, and downstream target gene expression of the NF-κB pathway.
