摘要:

目的:采用超高效液相色谱-四极杆-Orbitrap高分辨串联质谱(UPLC-Q-Exactive MS/MS)技术系统分析和解生发汤(HJSFT)的化学成分、入血原型成分及代谢产物,探讨其作用机制的物质基础。方法:采用Waters ACQUITY UPLC HSS T3色谱柱及Q-Exactive HFX高分辨质谱系统,对和解生发汤水煎剂及给药后小鼠血清进行化学成分与代谢产物检测与解析,结合特征碎片、文献及数据库比对,确定入血原型成分及代谢类型。结果:共鉴定出160种化学成分,其中入血原型成分33种,代谢产物9种。主要包括黄酮类、苯丙素类及萜类,代谢方式以葡萄糖醛酸化、硫酸化和甲基化为主。结论:和解生发汤经口给药后活性成分可吸收入血并发生二相代谢,其药效可能通过抗炎、抗氧化及免疫调节,实现对斑秃的治疗作用。

Abstract:

Objective: To systematically analyze the chemical components, blood-absorbed prototype components, and metabolites of Hejie Shengfa Tang (HJSFT) using UPLC-Q-Exactive MS/MS technology, and to explore the material basis of its mechanism of action. Methods: The chemical components and metabolites in the HJSFT water decoction and the serum of mice after administration were analyzed using a Waters ACQUITY UPLC HSS T3 column and a Q-Exactive HFX high-resolution mass spectrometry system. The blood-absorbed prototype components and metabolic types were identified by comparing characteristic fragments with literature and database information. Results: A total of 160 compounds were identified, including 33 blood-absorbed prototype components and 9 metabolites. These mainly included flavonoids, phenylpropanoids, and terpenes, with glucuronidation, sulfation, and methylation being the primary metabolic pathways. Conclusion: After oral administration, the active ingredients in Hejie Shengfa Tang can be absorbed into the blood and undergo phase metabolism. Its therapeutic effect on alopecia areata may be related to anti-inflammatory, antioxidant, and immunomodulatory actions.
