摘要:目的:优化助阳生精方煎煮工艺。方法:基于单因素试验结果,将液料比、提取时间和提取次数作为考察因素,以淫羊藿苷含量、黄芪甲苷含量和浸膏得率为评价指标,通过熵权法确定指标权重系数并计算综合评分。利用Box-Behnken响应面设计试验分析考察因素对综合评分的影响,根据综合评分优选最佳煎煮工艺并验证。结果:助阳生精方最佳煎煮工艺为液料比18∶1,提取1.4 h,提取2次。3次验证试验显示,淫羊藿苷、黄芪甲苷的平均含量分别为2.811 mg/g、0.404 mg/g,平均浸膏得率为32.716%,平均综合评分为97.92分,与预测值的相对误差均小于2%。结论:熵权法结合Box-Behnken响应面法优选的煎煮工艺稳定可行,可为助阳生精方的进一步开发研究提供试验基础。

Abstract:
Objective: To optimize the decoction process of the Zhuyang Shengjing Formula. Methods: Based on single factor experiments, the liquid-to-material ratio, extraction time, and number of extraction cycles were used as investigation factors, with the contents of icariin, astragaloside Ⅳ and the extract yield as evaluation indicators. The weight coefficients of the indicators were determined by the entropy weight method, and the comprehensive score was calculated accordingly. The Box-Behnken response surface design was employed to analyze the effects of the investigation factors on the comprehensive score. Based on the comprehensive score, the optimal decoction process was selected and validated. Results: The optimal decoction process for Zhuyang Shengjing Formula is a liquid to material ratio of 18:1, extraction for 1.4 hours, and extraction twice. The average contents of icariin and astragaloside Ⅳ, along with the extract yield, from three batches of validation experiments were 2.811 mg/g, 0.404 mg/g, and 32.716%, respectively. The average comprehensive score was 97.92, and the relative errors between these actual values and the predicted values were all less than 2%. Conclusion: The decoction process optimized by the entropy weight method combined with the Box-Behnken response surface method is stable and reliable, which can provide experimental basis for the further development and research of the Zhuyang Shengjing Formula.
