苓泽合剂对痛风性关节炎模型大鼠踝关节滑膜组织中MMP-1、LP-PLA2和ColⅡ的影响

摘要:目的:探讨苓泽合剂对痛风性关节炎模型大鼠踝关节滑膜组织中MMP-1LP-PLA2ColⅡ的影响。方法:将84SPF级健康雄性SD大鼠随机分为正常组、模型组、苓泽合剂低剂量组、苓泽合剂中剂量组、苓泽合剂高剂量组、依托考昔组和痛风定组,每组12只。除正常组外其余各组均采用3%氧嗪酸钾溶液腹腔注射结合尿酸钠(MSU)关节腔注射建立GA大鼠模型,造模成功后开始药物治疗,药物剂量跟随大鼠体质量等比例加减,干预一段时间后分别于造模前及造模4 h24 h48 h72 h后测量大鼠踝关节肿胀度,观察和检测各组大鼠模型踝关节滑膜组织病理学形态变化和MMP-1LP-PLA2和Ⅱ型胶原表达情况并分析其相关性。结果:与正常组比较,模型组大鼠各时间节点关节肿胀度均升高,大鼠模型关节滑膜组织中MMP-1表达升高,LP-PLA2表达升高、Ⅱ型胶原表达降低,Ⅱ型胶原含量明显降低(P0.001)。与模型组比较,苓泽合剂组能减轻大鼠关节肿胀度、降低MMP-1表达、降低LP-PLA2表达、增加Ⅱ型胶原表达。结论:苓泽合剂对急性痛风性关节炎MSU结晶沉积导致的炎症反应及软骨损伤有较好的防治作用。

Abstract:

Objective: To investigate the effects of Lingze mixture on MMP-1, LP-PLA2 and Colin the synovium of ankle in rats with gouty arthritis. Methods: Totally 84 healthy male SD rats with SPF grade were randomly divided into normal group, model group, Lingze mixture low-dose group, Lingze mixture medium-dose group, Lingze mixture high-dose group, etoricoxib group and tougfengding (痛风定) group, with 12 rats in each group. With the exception of the normal group, a gouty arthritis rat model was established in all other groups via intraperitoneal injection of a 3% potassium oxonate solution combined with intra-articular injection of monosodium urate (MSU). After successful modeling, drug treatment was started, and the drug dose was increased or decreased in proportion to the weight and mass of the rats. After intervention for a period of timethe ankle swelling degree of the rats was measured before and after 4 h, 24 h, 48 h and 72 hrespectively. The histopathological changes of ankle synovium and the expression of MMP-1, LP-PLA2 and type collagen in each group were observed and analyzed. Results: Compared with the normal group, the joint swelling degree of rats in the model group was increased at all time points, and the expression of MMP-1 and LP-PLA2 in the synovial tissue of the rat model joints was increased, while the expression of type collagen and serum content of type collagen was significantly decreased (P0.001). Compared with model groupLingze mixture group could reduce joint swelling, decrease MMP-1 and LP-PLA2 expression and increase type collagen expression. Conclusion: Lingze mixture can prevent and cure inflammation and cartilage damage in acute gouty arthritis caused by MSU crystal deposition.
