益气养血方通过激活CD4+T细胞抑制食管癌增殖的机制研究*

摘要:目的:探讨益气养血方通过激活CD4+T淋巴细胞杀伤作用抑制食管癌细胞增殖的机制。方法:采用ECA109人食管癌细胞和CD4+T淋巴细胞共培养模型,加入0.5 μg/mL益气养血方进行干预,通过细胞计数(CCK-8)实验、流式细胞术、酶联免疫吸附试验(ELISA)和免疫印迹(Western blotting)等方法,观察益气养血方对食管癌细胞增殖、凋亡及磷脂酰肌醇-3-激酶/蛋白激酶BPI3K/Akt)信号通路的影响。结果:CCK-8和流式细胞术表明益气养血方能进一步增强CD4+T细胞的杀伤作用,显著降低食管癌细胞的存活率(P<0.05)。ELISA结果显示,在共培养体系中益气养血方能够显著上调γ干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)和乳酸脱氢酶(LDH)的表达水平(P<0.05)。Western blotting结果显示,在共培养体系中益气养血方能够显著下调食管癌细胞中p-PI3Kp-Akt1的表达水平,抑制PI3K/Akt信号通路的激活(P<0.05)。结论:益气养血方能通过激活CD4+T淋巴细胞的杀伤作用,抑制食管癌细胞增殖,其机制可能与抑制PI3K/Akt信号通路的激活有关。

Abstract:

Objective: To explore the mechanism of Yiqi Yangxue decoction inhibiting the proliferation of esophageal cancer cells by activating the killing effect of CD4+T lymphocytes. Methods: The co-culture model of ECA109 human esophageal cancer cells and CD4+T lymphocytes was used, and Yiqi Yangxue decoction (0.5 μg/mL) was added for intervention. Through cell counting kit-8 (CCK-8) experiments, flow cytometry, enzyme-linked immunosorbent assay (ELISA), and Western blotting, the effects of Yiqi Yangxue decoction on esophageal cancer cell proliferation, apoptosis, and the phosphatidylinositol-3-kinase/protein kinase B (PI3K/Akt) signaling pathway were observed. Results: CCK-8 and flow cytometry showed that Yiqi Yangxue decoction could further enhance the killing effect of CD4+T cells and significantly reduce the survival rate of esophageal cancer cells (P<0.05). ELISA results showed that Yiqi Yangxue decoction could significantly up-regulate the expression levels of IFN-γ, TNF-α, and LDH in the co-culture system (P<0.05). Western blotting results showed that Yiqi Yangxue decoction could significantly down-regulate the expression levels of p-PI3K and p-Akt1 in esophageal cancer cells in the co-culture system, and inhibit the activation of the PI3K/Akt signaling pathway (P<0.05). Conclusion: Yiqi Yangxue decoction can inhibit the proliferation of esophageal cancer cells by activating the killing effect of CD4+T lymphocytes, and its mechanism may be related to the inhibition of the activation of the PI3K/Akt signaling pathway.
